Now, should this happen ? Should I be able to lock the reel off 100% ?

Strictly speaking, no, not on any reel I've ever owned. Practically
speaking, you can usually screw the drag tight enough that the line
snaps before it gives. With 50lb line, though, perhaps not. I'd be
surprised if you can get a small spinning reel with a tight enough drag
to resist that.

How hard are you pulling the line? You have found and tightened the drag
knob?
